okay so good morning everybody uh thank you so much uh for all
00:00:06
for coming to visit us at e. p. f. l. campus today
00:00:09
so all be telling you about uh the work that we've been
00:00:12
doing in our lab uh related to competition operating design
00:00:16
and uh the potential applications uh for this methodologies in by madison
00:00:20
applications so if uh many of you are aware um
00:00:25
most of the proteins we know in fact a evolutionary as history meaning that um they were
00:00:30
exposed to particular environment or they were not involved in a particular context of an organism
00:00:36
about what we can do these days um if in fact i have a
00:00:40
protein design tools which in which in this case will be computational tools
00:00:44
um to in fact change at the street uh of these proteins
00:00:49
and what i say about changing the each of these proteins is essentially to manipulate them in a way that they can perform
00:00:54
new functions for us and the way we do this uh is in fact uh we can draw parallel maybe with um
00:01:01
molecular sculpture so essentially we start from protein structures and we
00:01:05
try to find new sequences that that fit to structures
00:01:08
and ultimately the aim here is really good she's a new different functions and
00:01:13
so and uh in the next slide some just gonna go through a couple of of units of projects that are lab is interested in
00:01:20
so one of the first things that we've done uh using these techniques in order to generate novel proteins
00:01:26
was to apply this and what we called the top focused vaccines and so here what
00:01:31
you see uh on the left uh is essentially the uh a structural motif
00:01:36
that are low protein displays and that our immune system can recognise and um
00:01:41
make antibodies to target at this this motif so once we put this motif into
00:01:46
what difference interpret in that we design in the computer we were then
00:01:49
uh able to use the synthetic vaccines in a animal
00:01:53
intersections and in fact um verify that we
00:01:56
could elicit i'm controlling and somebody's against intense uh
00:01:59
make uh the principal components for active vaccines
00:02:03
so up until to day um this was done in in the particular system which is called the respiratory such a virus
00:02:09
for which there still no backs in approving this is in an area of research that we are intensively working on
00:02:16
so on the next example just to give you again uh another showcase of what this technologies can do for us
00:02:21
we're also working this domain of on a anti cancer drugs and so here what we
00:02:25
did was essentially to design a new protein which the target uh an essential factor
00:02:30
in in cells which could in it it onto my growth in this essentially the data
00:02:34
that i'm showing you here which are all was also data collected in people
00:02:38
and so but what i want you to in fact appreciate up from the
00:02:40
slide is that by having and this ability to to manipulate the structures
00:02:45
with an essentially very similar structures one another uh using essentially very close proteins to each other
00:02:53
we can re purpose them for completely different applications one of them being an anti too much rock the other one being a vaccine or
00:03:02
um one other aspect that we are also working on
00:03:05
is in um designing a molecular component read
00:03:09
which we can um control a cell activities and so here the principle is is essentially very simple
00:03:15
let's say that you have to proteins in p. that come together they will yield i'm an active solar activity
00:03:21
um in the presence of the small molecule this will break the to produce a come together and
00:03:26
this will render an inactive so so right now we're still working this a very um
00:03:32
for very sometimes but um uh uh for now we have in future
00:03:37
proof of concept that we can in fact make this happen
00:03:40
and this will also have a rapid translation applications i'm into parties out there please
00:03:45
so i'll ultimately ah what it but the message that i want to convey today is really that uh
00:03:51
this uh cutting edge pretty mentioning techniques are in
00:03:54
fact enabling interest formative for um biomedical applications
00:03:58
uh and so here is just a list of um that uh applications that we are working right now obviously we're open to other ideas
00:04:05
um and at the same time i will obviously would like to thank the people that actually do the
00:04:09
work in my lap and also the terms funding that we have obtained for the last years
